awk gsub with variables

Hello,

I’m trying to substitute a string with leading zero for all the records except the trailer record using awk command and with variables. The input file test_med1.txt has data like below

1234ABC………………………9200…………LF
456789………………………..0401…………LF

And I want to replace 9200 with 09200 and 0401 with 00401

1234ABC………………………09200…………LF
456789………………………..00401…………LF

I tried below command but not successfull

Command :

a= cat test_med1.txt | awk ‘!/TLR/’ | cut -c 65-84 b= `cat test_med1.txt | awk ‘!/TLR/’ | cut -c 65-84 | awk ‘{$0=0$0;print}’`; awk -v rvar=${a} -v svar=${b} ‘{gsub(rvar,svar); print}’ test_med1.txt

Error:

awk: 0602-533 Cannot find or open file 07803.
The source line number is 1.

Please let me know where the command in failing and if it is complete wrong then let me know how to get above output using awk and gsub command

Idera Pharmaceuticals Reports Fourth Quarter and Full Year 2012 Financial Results and Provides Update on Autoimmune Disease Program

CAMBRIDGE, Mass.–(BUSINESS WIRE)– Idera Pharmaceuticals, Inc. (NAS: IDRA) today reported its financial and operational results for the fourth quarter and year ended December 31, 2012.

“During the past year, Idera has demonstrated promising clinical activity for the use of Toll-like Receptor antagonists as a novel approach for the potential treatment of autoimmune diseases,” said Sudhir Agrawal, D.Phil., Chairman and Chief Executive Officer. “We recently completed dosing in the escalating single-dose portion of our Phase 1 study of IMO-8400, an antagonist of TLRs 7, 8 and 9. In this portion of the trial, IMO-8400 was well-tolerated and showed TLR target engagement at three dose levels in healthy subjects. These results build on the results of our Phase 2 clinical trial of IMO-3100, an antagonist of TLRs 7 and 9, in patients with plaque psoriasis, which we believe provided clinical proof of concept of our approach of targeting specific TLRs for the treatment of psoriasis and other autoimmune and inflammatory diseases.”

Dr. Agrawal continued, “Our next step is to conduct a Phase 2 clinical trial in patients with psoriasis with an extended treatment period of up to 12 weeks. Based on our evaluation of the comparative profiles of IMO-3100 and IMO-8400, including the engagement of TLR8 by IMO-8400, Idera has determined to conduct this Phase 2 trial with IMO-8400. Subject to successful completion of our ongoing multiple dose Phase 1 trial of IMO-8400 and to obtaining the necessary financing to conduct this trial, we expect to initiate this Phase 2 clinical trial in the second quarter of 2013.”

Idera Announces Presentation at AAD Annual Meeting Demonstrating Toll-like Receptor Antagonists Normalize Expression of Inflammatory Genes in Psoriasis Model

–Phase 2 Data of IMO-3100 in Psoriasis to be Presented at IID Meeting 2013–

CAMBRIDGE, Mass.–(BUSINESS WIRE)– Idera Pharmaceuticals, Inc. (NAS: IDRA) today announced the presentation of new data showing that its selective Toll-like receptor antagonists, IMO-3100 and IMO-8400, normalized the gene expression of important cytokines in a preclinical study of skin inflammation that is commonly used as a model of psoriasis. Notably, genes related to the production of key mediators of psoriasis including Interleukin (IL) -17, IL-6, IL-12/23, IL-1, IL-21 Receptor, and INF-gamma were restored toward normal levels. The data were presented by James G. Krueger, M.D., Ph.D, of The Rockefeller University, at the Late-Breaking Research Symposium on March 2nd, 2013, during the American Academy of Dermatology Annual Meeting.

“Our studies show that treatment with IMO-3100 and IMO-8400 reduces disease-associated expression of the IL-17 and IL-23 inflammatory cascade in the mouse IL-23-induced skin inflammation model,” said Dr. James Krueger. “Genes that play a mediating role in psoriasis were returned toward normal levels with both compounds; and the inclusion of TLR8 activity with IMO-8400 was additive to the effect on gene expression that we observed with the TLR7 and TLR9 antagonist compound. We are now extending this work, with the analysis of gene expression patterns in skin biopsies from patients treated with a TLR antagonist in a Phase 2 clinical trial.”
